Under a magnification of 1000X, this Ziehl-Neelsen-stained tissue specimen, revealed the presence of tubercle bacilli, Mycobacterium tuberculosis, the bacterium responsible for causing tuberculosis. Ziehl-Neelsen technique is used when staining acid-fast bacteria, implementing a carbol-fuchsin solution, then decolorizing in acid alcohol, and counterstaining with methylene blue. Mycobacteria have waxy-coated cell walls, which prevents effective Gram-staining.
