This was an artist’s representation of a photomicrograph, which had been processed using the Ziehl-Neelsen staining technique, revealing the presence of Mycobacterium tuberculosis bacteria in this sputum smear. The Ziehl-Neelsen technique is used for staining acid-fast bacteria, implementing a carbol-fuchsin solution, while decolorizing in acid alcohol, and counterstaining with methylene blue. Mycobacteria have waxy coated cell walls, which prevents Gram-staining.
