This 1959 photomicrograph depicted a mixed culture of microorganisms, which had been histochemically processed using the fluorescent antibody (FA) staining method. In the field of view, one can see both rod shaped bacilli, and small, ovoid shaped cocci bacteria. The FA test is accomplished, by molecularly attaching a fluorescent dye to an antibody with an affinity, or attraction to an antigen, located on the surface of a microorganism. Once the antibody-dye complex has attached to the bacterium, the sample may then be observed under ultraviolet illumination, which will cause the fluorescent dye to emit light.
