This Petri dish culture plate contained Thayer-Martin growth medium, to which the reagent, N,N,N',N'-tetramethyl-p-phenylenediamine (TMPD) had been added. The plate was then inoculated with a bacterial culture obtained from a cervical specimen, harvested from a suspected gonorrhea patient. As you can see, after having been incubated for an unspecified length of time, the plate produced numerous, tiny, purple-black bacterial colonies, indicative of a positive oxidase test. Neisseria gonorrhoeae bacteria are oxidase-positive, possessing the enzyme cytochrome c oxidase, and are therefore, able to oxidize TMPD, producing this purple-black coloration.
